An Experimental Study on the Enhanced Cytolytic Effects of Natural Killer Cells by Interleukin2




Abstract
Culturing of spleen cells of mice (C57BL/6) with IL-2 induces NK cell activity. NK cell which is a nonspecific killer cell kills various target cells, like tumor cells or virally infected cells, and exhibit heterogeneity with respect to their
target
cell specificities. Optimizing the concentration of IL-2 will correspondingly enhance NK cell activity which can be represented by target cell lysis.
In this experiment, murine spleen cells were cultured without IL-2 as a control grouup. On the other arm, cells with various concentrations of IL-2, gradually from 1.6, 3.1, 6.25, 12.5 to 25 IU/ml, were placed into each wells of tissue culture
plate.
The cell line of Moloney leukemia virus induced lymphoma(YAC-1) was used as a target cell and te degree of target cell lysis was measured in the 4 hours 51Cr-release assay.
@ES The result of this experiment was as follows:
@EN 1) The Natural killer activity started from lst day of culture, and peaked on 2nd day of culture in the wells with relatively higher (Effector cell(E)/Target cell(T) ratio 100 and 50) concentrations of effector cell(p<0.001).
2) At 3rd day of the culture, wells with the concentration of IL-2 less than 6.25 IU/ml and E/T ratio of 12.5, negative % specific lysis also emerged in the well swith the concentration of IL-2 less than 3.1 IU/ml and E/T of 25.
3) The higher concentration of IL-2, the greater target cell lysis could be observed specifically in the wells with relatively higher concentrations(E/T ratio 100 and 50) of effector cell(p<0.0001). but there was no statistically significant
correlation between the concentrations of IL-2 and the degree of target cell lysis in the wells with relatively lower concentrations(E/T ratio 25 and 12.5) of effector cell.
